Description

GOT2 Mouse Recombinant produced in E.Coli is a single, non-glycosylated polypeptide chain containing 422 amino acids (30-430 aa) and having a molecular mass of 46.8kDa.
GOT2 is fused to a 21 amino acid His tag at N-terminus and purified by proprietary chromatographic techniques.

Product Specs

Introduction
GOT2 is a pyridoxal phosphate-dependent enzyme found in both the cytoplasm and the inner mitochondrial membrane, existing as GOT1 and GOT2, respectively. GOT2 is involved in amino acid metabolism as well as the urea and tricarboxylic acid cycles. Both GOT1 and GOT2 are homodimeric and share close homology.
Description
Recombinant Mouse GOT2, expressed in E. coli, is a single, non-glycosylated polypeptide chain containing 422 amino acids (residues 30-430). The protein has a molecular mass of 46.8 kDa. For purification purposes, a 21 amino acid His tag is fused to the N-terminus. The protein is purified using proprietary chromatographic techniques.
Physical Appearance
Sterile Filtered colorless solution
Formulation
The GOT2 solution is provided at a concentration of 0.5 mg/ml in a buffer composed of 10% Glycerol and Phosphate Buffered Saline (pH 7.4).
Stability
For short-term storage (2-4 weeks), store at 4°C. For longer storage, freeze at -20°C. Adding a carrier protein (0.1% HSA or BSA) is recommended for extended storage. Avoid repeated freeze-thaw cycles.
Purity
Purity is determined to be greater than 90.0% by SDS-PAGE analysis.
Biological Activity
The specific activity is greater than 20 units/mg. This is measured as the amount of enzyme required to convert 1 μmole of alpha-ketoglutarate to L-Glutamate per minute at pH 8.0 and 25°C.

Product Science Overview

Introduction

Glutamic-Oxaloacetic Transaminase 2 (GOT2), also known as Aspartate Aminotransferase 2 (AST2), is a pyridoxal phosphate-dependent enzyme that plays a crucial role in amino acid metabolism and the urea and tricarboxylic acid cycles . This enzyme exists in both cytoplasmic and mitochondrial forms, with GOT2 being the mitochondrial variant .

Structure and Function

GOT2 is a homodimeric enzyme, meaning it consists of two identical subunits. Each subunit contains several domains, including a large domain that binds pyridoxal phosphate, a small domain, an NH2-terminal arm, and a bridge formed by specific residues . The enzyme catalyzes the transamination of oxaloacetate to aspartate, a key reaction in the malate-aspartate shuttle, which is essential for transferring reducing equivalents across the mitochondrial membrane .

Role in Metabolism

GOT2 is integral to the malate-aspartate shuttle, which facilitates the transfer of NADH from the cytosol to the mitochondria for energy production . This shuttle is vital for maintaining the NAD+/NADH balance within cells, which is crucial for various metabolic processes, including glycolysis and the electron transport chain .

Recombinant GOT2 in Research

Recombinant GOT2, particularly the mouse variant, is produced in E. coli and is used extensively in research. The recombinant protein is a single, non-glycosylated polypeptide chain containing 422 amino acids and has a molecular mass of approximately 46.8 kDa . It is often fused to a His-tag at the N-terminus to facilitate purification .

Applications in Biomedical Research

Recombinant GOT2 is used in various biochemical assays to study its role in amino acid metabolism and its involvement in diseases such as developmental and epileptic encephalopathy and pheochromocytoma . Additionally, GOT2’s role in the malate-aspartate shuttle makes it a target for research into metabolic disorders and potential therapeutic interventions .
