Description

CHGA produced in Sf9 Baculovirus cells is a single, glycosylated polypeptide chain (19-457 a.a.) and fused to a 6 aa His Tag at C-terminus containing a total of 448 amino acids and having a molecular mass of 50kDa.CHGA shows multiple bands between 50-70kDa on SDS-PAGE, reducing conditions and purified by proprietary chromatographic techniques.

Product Specs

Introduction
Chromogranin-A Isoform 1 Preproprotein, also known as CHGA, belongs to the chromogranin/secretogranin family of neuroendocrine secretory proteins. It serves as a precursor to various enzymes, including pancreastatin, catestatin, vasostatin-1, vasostatin-2, and parastatin. CHGA functions as a negative regulator of neuroendocrine activity in autocrine or nearby cells (paracrine). In pancreatic islet beta cells, CHGA promotes the production of secretory granules containing insulin.
Description
Produced in Sf9 Baculovirus cells, CHGA is a single, glycosylated polypeptide chain encompassing amino acids 19-457. It is fused to a 6-amino acid His Tag at the C-terminus, resulting in a protein with a total of 448 amino acids and a molecular weight of 50 kDa. Under reducing conditions on SDS-PAGE, CHGA displays multiple bands between 50-70 kDa. It is purified using proprietary chromatographic techniques.
Physical Appearance
Sterile Filtered colorless solution.
Formulation
CHGA protein solution is provided at a concentration of 0.25 mg/ml in Phosphate Buffered Saline (pH 7.4) containing 20% glycerol and 0.1 mM PMSF.
Stability
For short-term storage (up to 2-4 weeks), store at 4°C. For extended storage, freeze at -20°C. Adding a carrier protein (0.1% HSA or BSA) is recommended for long-term storage. Avoid repeated freeze-thaw cycles.
Purity
Purity is determined to be greater than 85.0% by SDS-PAGE analysis.

Product Science Overview

Structure and Function

CgA is a precursor to numerous bioactive peptides and plays a crucial role in the regulation of neuroendocrine functions. It acts as a negative regulator of the neuroendocrine activity of autocrine or nearby cells (paracrine) . The protein is involved in the formation of secretory granules that contain insulin in pancreatic islet beta cells .

Recombinant Production in Sf9 Cells

The recombinant form of CgA is produced in Sf9 Baculovirus cells. This recombinant protein is a single, glycosylated polypeptide chain consisting of 448 amino acids, including a 6 amino acid His Tag at the C-terminus . The molecular mass of this recombinant protein is approximately 50 kDa, and it shows multiple bands between 50-70 kDa on SDS-PAGE under reducing conditions .

Biomedical Applications

CgA and its derived peptides have significant biomedical applications. They are used as biomarkers for various diseases, including neuroendocrine tumors . The protein’s role in the neuroendocrine, immune, and digestive systems makes it a valuable target for translational and precision medicine .

Storage and Stability

The recombinant CgA protein solution is formulated in Phosphate Buffered Saline (pH 7.4) with 20% glycerol and 0.1mM PMSF . For short-term storage, it should be kept at 4°C if used within 2-4 weeks. For long-term storage, it is recommended to freeze the protein at -20°C with a carrier protein (0.1% HSA or BSA) to avoid multiple freeze-thaw cycles .
