Introduction
Uteroglobin (SCGB1A1), a member of the Secretoglobin (SCGBs) family, is a versatile protein known for its anti-inflammatory and anti-tumorigenic properties. It achieves this by binding to small hydrophobic molecules, such as phospholipids and prostaglandins. Uteroglobin plays a crucial role in various biological processes, including inflammation suppression, phospholipase A2 inhibition, and the sequestration of hydrophobic ligands. Primarily produced by Clara cells, the non-ciliated, non-mucous secretory cells found in lung bronchioles, SCGB1A1 is also expressed by other epithelia that interact with the external environment. Beyond its role in sequestering pro-inflammatory mediators and carcinogens, Uteroglobin is involved in inhibiting cell migration and invasion, platelet aggregation, and T cell differentiation. Notably, defects in the SCGB1A1 gene are linked to an increased susceptibility to asthma.
Description
Recombinant Human Uteroglobin, expressed in E. coli, is a single, non-glycosylated polypeptide chain consisting of 80 amino acids. It includes a 10 amino acid N-terminal His tag, resulting in a calculated molecular mass of 9.2kDa.
Physical Appearance
White, lyophilized powder after filtration.
Formulation
The protein solution, at a concentration of 0.5 mg/ml in 0.05M phosphate buffer with 0.075M NaCl at pH 7.4, is filtered through a 0.4 μm filter and then lyophilized.
Solubility
To prepare a working stock solution, add deionized water to the lyophilized pellet to achieve a concentration of approximately 0.5 mg/ml. Allow the pellet to dissolve completely. Note that SCGB1A1 is not sterile. Before using in cell culture, filter the product through an appropriate sterile filter.
Stability
Store the lyophilized protein at -20°C. After reconstitution, aliquot the product to minimize freeze-thaw cycles. The reconstituted protein remains stable at 4°C for a limited period and shows no significant change for up to two weeks at this temperature.
Purity
The purity is determined to be greater than 90.0% using SDS-PAGE analysis.
