Product Specs

Introduction
Interleukin 7 (IL-7) is a crucial cytokine for the development of B and T cells. In conjunction with hepatocyte growth factor (HGF), it forms a heterodimer that stimulates the growth of pre-pro-B cells. Moreover, IL-7 acts as a cofactor during early T cell development, facilitating the V(D)J rearrangement of the T cell receptor beta (TCRB). Produced by intestinal epithelial and epithelial goblet cells, IL-7 might also regulate intestinal mucosal lymphocytes. Studies involving knockout mice have demonstrated the indispensable role of IL-7 in lymphoid cell survival.
Description
Produced in Sf9 insect cells using a baculovirus expression system, our recombinant human IL-7 is a single, glycosylated polypeptide chain. It encompasses amino acids 26-177, resulting in a protein of 161 amino acids with a molecular weight of 18.4 kDa. A 6-amino acid Histidine tag is present at the C-terminus to facilitate purification. The protein undergoes purification using proprietary chromatographic techniques.
Physical Appearance
Clear, colorless, and sterile-filtered solution.
Formulation
The IL-7 protein is supplied in a solution at a concentration of 0.5 mg/ml. The solution is composed of Phosphate Buffered Saline (PBS) at a pH of 7.4 and contains 10% glycerol as a stabilizing agent.
Stability
For short-term storage (up to four weeks), the IL-7 protein should be stored at 4°C. For extended storage, it is recommended to store the protein in aliquots at -20°C. The addition of a carrier protein, such as 0.1% HSA or BSA, is recommended for long-term storage to maintain protein stability. Avoid repeated freeze-thaw cycles to prevent protein degradation.
Purity
The purity of the IL-7 protein is greater than 90%, as determined by SDS-PAGE analysis.
Synonyms
Interleukin 7, IL-7, Interleukin-7.

Product Science Overview

Introduction

Interleukin-7 (IL-7) is a crucial cytokine involved in the development and homeostasis of the immune system. It plays a significant role in the proliferation and survival of T and B cells, making it essential for maintaining a functional immune response . Recombinant human IL-7, produced using the sf9 insect cell expression system, is widely used in research and therapeutic applications due to its high purity and biological activity .

Production and Preparation

Recombinant human IL-7 is typically produced using the baculovirus expression vector system (BEVS) in sf9 insect cells. This method involves the following steps:

  1. Gene Cloning: The human IL-7 gene is cloned into a baculovirus transfer vector.
  2. Transfection: The recombinant baculovirus is used to transfect sf9 insect cells.
  3. Protein Expression: The transfected sf9 cells express the IL-7 protein.
  4. Purification: The IL-7 protein is purified using chromatography techniques to achieve high purity and biological activity .
Biological Functions

IL-7 is a member of the type I cytokine family and is primarily produced by non-hematopoietic stromal cells in the bone marrow, thymus, and other lymphoid organs . It acts as a growth factor for T cells and B cells, promoting their development, survival, and homeostasis. IL-7 also plays a critical role in the maintenance of naive and memory T cells by preventing apoptosis .

Mechanism of Action

IL-7 exerts its effects through a heterodimeric receptor composed of the IL-7 receptor alpha chain (IL-7Rα) and the common gamma chain (γc), which is shared with other cytokine receptors such as IL-2, IL-4, IL-9, IL-15, and IL-21 . Binding of IL-7 to its receptor activates several intracellular signaling pathways, including the JAK-STAT and PI3K-AKT pathways, leading to cell proliferation, survival, and differentiation .

Clinical Applications

Recombinant human IL-7 has shown promise in various clinical applications, particularly in immune reconstitution and cancer immunotherapy. Clinical trials have demonstrated its safety and efficacy in enhancing immune responses in patients with immunodeficiency disorders and certain types of cancer . Additionally, IL-7 is being investigated for its potential to improve vaccine responses and treat chronic infections .
