IL-19 is preferentially expressed in monocytes, macrophages, and T and B lymphocytes . It is produced by B cells and monocytes and binds to the interleukin 20 receptor complex to activate STAT3 signaling . The production of IL-19 is up-regulated in resting monocytes following stimulation with granulocyte-macrophage colony-stimulating factor (GM-CSF) or lipopolysaccharide (LPS) .
IL-19 plays a significant role in the immune response by promoting T helper 2 (Th2) cell-mediated immune responses . It induces the expression of IL-6 and tumor necrosis factor alpha (TNF-α) in monocytes . IL-19 initiates the JAK-STAT signaling pathway, which activates genes and creates mRNA sequences that are translated into proteins with downstream effector functions . The signaling involves the IL-20 dimer receptor complexes that bind the IL-19 ligand, Janus kinases (JAKs), and the signal transducer and activator of transcription 3 (STAT3) .
Recombinant IL-19 (Human Recombinant, HEK) is produced in HEK 293 cells. It is typically used in research settings to study its effects on immune responses and its potential therapeutic applications . The recombinant form of IL-19 is available in various purities and formulations, and it is often lyophilized from a sterile filtered solution containing trifluoroacetic acid (TFA) .
IL-19 has broad functions across inflammation, cell development, viral responses, and lipid metabolism . As an immunosuppressive cytokine, IL-19 promotes an anti-inflammatory lymphocyte phenotype, dampens the Th1 T-cell response, and increases IL-10 expression in peripheral blood mononuclear cells (PBMC) . It also inhibits the production of immunoglobulin G (IgG) from B cells .
