Introduction
CD40L, also known as CD154, is a surface protein found on T cells. It plays a crucial role in the immune response by interacting with CD40 on B cells. This interaction is vital for B cell activation, proliferation, and differentiation, leading to antibody production. Additionally, CD40L can stimulate other immune cells like monocytes, triggering them to release signaling molecules (cytokines) and enhance their ability to eliminate tumor cells. It can also promote the proliferation of activated T cells and induce the production of important cytokines like IFN-gamma, TNF-alpha, and IL-2.
Description
Recombinant Mouse sCD40, produced in E. coli, is a non-glycosylated polypeptide chain comprising 149 amino acids. With a molecular weight of 16409 Daltons, this protein represents the soluble form of CD40. It undergoes purification using proprietary chromatographic techniques to ensure its high quality.

Formulation
Lyophilized from a sterile solution containing 1mg/ml sCD40 and 10mM Sodium Phosphate buffer at pH 7.5.
Solubility
For reconstitution, it is recommended to dissolve the lyophilized sCD40 in sterile 18MΩ-cm H2O to a concentration of at least 100 µg/ml. This solution can be further diluted in other aqueous solutions as needed.
Stability
Lyophilized sCD40 remains stable at room temperature for up to 3 weeks; however, for long-term storage, it is recommended to store it desiccated below -18°C. After reconstitution, CD154 should be stored at 4°C and used within 2-7 days. For extended storage, adding a carrier protein (0.1% HSA or BSA) is advisable. Avoid repeated freeze-thaw cycles.
Purity
The purity of sCD40 is greater than 98.0%, as determined by two methods: (a) Reverse-Phase High-Performance Liquid Chromatography (RP-HPLC) and (b) Sodium Dodecyl Sulfate-Polyacrylamide Gel Electrophoresis (SDS-PAGE).
Biological Activity
The biological activity of sCD40 was assessed by its ability to induce MIP-1 alpha and TNF-alpha production in mouse splenocytes. The ED50, which represents the concentration at which 50% of the maximal response is observed, was determined to be 0.1 µg/ml. This corresponds to a Specific Activity of 10,000 IU/mg.
