Product Specs

Introduction

Seven known coronaviruses are known to infect humans, including OC43. The others are CoV-229E, CoV-NL63, CoV-HKU1, MERS-CoV, the original SARS-CoV1, and SARS-CoV-2 (Covid 19). CoV-OC43, human alpha-coronavirus 229E and NL63, beta-coronavirus and HCoV-HKU1 are responsible for the common cold. Like other betacoronaviruses, CoV-OC43 possesses an additional, shorter spike protein known as hemagglutinin esterase. Human coronavirus OC43 is a member of the beta-coronavirus species. These viruses are enveloped, positive-sense, single-stranded RNA viruses that gain entry into their host cell by binding to the N-acetyl-9-O-acetylneuraminic acid receptor.

Description

Recombinant Human Coronavirus OC43 Nucleoprotein is a full-length protein expressed in E. coli. The predicted signal peptide of the first 30 amino acids is not present in this recombinant protein. The protein migrates at 50kDa.
The CoV OC43 protein was designed with a C-terminal 6xHis tag and is purified using a proprietary chromatographic technique.

Product Science Overview

Introduction

Human coronavirus OC43 (HCoV-OC43) is one of the four coronaviruses that commonly cause mild respiratory infections, often referred to as the common cold. It belongs to the Betacoronavirus genus, which also includes the more severe SARS-CoV and MERS-CoV . HCoV-OC43 was first isolated in the 1960s from patients with upper respiratory tract infections .

Structure and Function of Nucleoprotein

The nucleoprotein (N) of HCoV-OC43 plays a crucial role in the virus’s life cycle. It is responsible for binding to the viral RNA genome, forming the ribonucleoprotein complex, and protecting the RNA from degradation. Additionally, the nucleoprotein is involved in the replication and transcription of the viral genome .

Recombinant Nucleoprotein

Recombinant nucleoprotein refers to the nucleoprotein that has been produced using recombinant DNA technology. This involves inserting the gene encoding the nucleoprotein into an expression system, such as bacteria or yeast, to produce the protein in large quantities. Recombinant nucleoproteins are valuable for research and diagnostic purposes, as they can be used to study the virus’s structure and function, develop diagnostic assays, and screen for potential antiviral drugs .

Immunological Significance

The nucleoprotein of HCoV-OC43 is a key target for the immune system. Studies have shown that the nucleoprotein contains multiple epitopes that are recognized by CD4+ T cells, which play a critical role in the immune response to viral infections . Some of these epitopes are specific to HCoV-OC43, while others are cross-reactive with other human coronaviruses, including SARS-CoV-2 . This cross-reactivity suggests that exposure to HCoV-OC43 may provide some level of immune protection against other coronaviruses.

Applications in Research and Diagnostics

Recombinant HCoV-OC43 nucleoprotein is widely used in research to study the virus’s biology and immune response. It is also used in the development of diagnostic assays, such as enzyme-linked immunosorbent assays (ELISAs), to detect antibodies against HCoV-OC43 in patient samples . These assays are important for understanding the epidemiology of HCoV-OC43 infections and for monitoring immune responses in vaccinated or infected individuals.
