Product Specs

Introduction
Peptidase Inhibitor 16 (PI16), a member of the CRISP family, is believed to inhibit serine proteases. It has been shown to interact with PSP94/MSMB. Expression is observed in the prostate, testis, ovary, and intestine. Notably, elevated levels of PI16 are found in the blood serum of individuals diagnosed with prostate cancer, suggesting its potential utility as a biomarker for monitoring patients following prostate cancer surgery.
Description
This recombinant human PI16 protein is produced in HEK293 cells. An 11-amino acid Flag tag is fused to the C-terminus of the protein. The resulting PI16 Flag-tagged fusion protein has a molecular weight of 45.7 kDa and consists of 426 amino acid residues. Purification is achieved using proprietary chromatographic techniques.
Physical Appearance
White, lyophilized powder after filtration.
Formulation
The product is provided as a lyophilized powder, having been sterile-filtered (0.4 μm) from a solution containing 0.5 mg/ml PI16 in 20mM Tris buffer (pH 7.5) and 50mM NaCl.
Solubility
To prepare a working stock solution, add deionized water to the lyophilized powder to a final concentration of 0.5 mg/ml. Allow the pellet to dissolve fully. Note: This product is not sterile. Prior to use in cell culture, it is essential to filter the reconstituted protein using an appropriate sterile filter.
Stability
The lyophilized PI16 human recombinant should be stored at -20°C. After reconstitution, aliquot the product to minimize freeze-thaw cycles. Reconstituted protein can be stored at 4°C for up to two weeks without observable degradation.
Purity

Purity exceeds 95.0% as determined by SDS-PAGE analysis.

Product Science Overview

Structure and Isoforms

PI16 has two known isoforms:

  1. Long Isoform: Comprising 463 amino acids with a molecular weight of approximately 49 kDa.
  2. Short Isoform: Comprising 270 amino acids with a molecular weight of approximately 30 kDa .
Expression and Localization

PI16 is expressed in various tissues, including the prostate, testis, ovary, and intestine. It is particularly concentrated in the sera of prostate cancer patients, suggesting its potential role as a biomarker for this disease .

Function

PI16 is a putative serine protease inhibitor. Protease inhibitors are essential for regulating protease activity, which is crucial for various physiological processes, including digestion, immune response, and blood coagulation . The exact biological function of PI16 is still under investigation, but its role in inhibiting serine proteases suggests it may be involved in modulating proteolytic activities in the tissues where it is expressed.

Recombinant Production

Recombinant Human PI16 is produced using various expression systems, including HEK293 cells and wheat germ. These systems ensure the correct folding and post-translational modifications necessary for the protein’s biological activity .

  • HEK293 Cells: This system is commonly used for producing recombinant proteins due to its ability to perform complex post-translational modifications. The recombinant PI16 produced in HEK293 cells is often tagged with a C-terminal Flag Tag for easy purification and detection .
  • Wheat Germ Expression System: This in vitro system preserves the correct conformational folding of the protein, which is crucial for its functionality. The recombinant PI16 produced in this system is tagged with GST at the N-terminal .
Applications

Recombinant PI16 is used in various research applications, including:

  • Enzyme-Linked Immunosorbent Assay (ELISA): For detecting and quantifying PI16 in biological samples.
  • Western Blotting (WB): For analyzing the expression of PI16 in different tissues.
  • Immunoaffinity Purification: For isolating PI16 from complex mixtures .
Clinical Relevance

PI16’s expression in prostate cancer patients’ sera highlights its potential as a prognostic marker for prostate cancer. Further research is needed to fully understand its role in cancer biology and its potential as a therapeutic target .
